Expression and cellular localisation of essential FIKKs within infected red blood cells. FIKK9.1,FIKK10.1 and FIKK10.2 were detected using anti-HA antibodies using IFA and were shown to localise to the RBC cytoplasm as punctate dots (FIKK9.1 and FIKK10.1) or both punctate and diffuse (FIKK10.2). Ring stage (left panel) and trophozoite/schizont stage parasites (right panel) were labelled with anti-HA antibodies (green) and PfSBP1 antibody (red). Nuclear staining using DAPI was in blue. The third column in each panel (except for FIKK9.5) is the merge of red and green channels and yellow areas represent regions of co-localisation. The fourth column in each panel (except for FIKK9.5) is the merge of red, green, and blue and the fifth column is the bright field image. FIKK9.1 and FIKK10.1 co-localized with PfSBP1, while FIKK10.2 partially co-localised with PfSBP1. FIKK3 was detected using anti-HA antibodies in IFA and shown to localize as dots adjacent to the nucleus specific to segmented schizonts and merozoites. Segmented schizonts were co-labelled with antibodies against PfRAMA (red). FIKK3 co-localised with PfRAMA, suggesting apical organelle localisation. FIKK9.5 was detected using anti-HA antibodies in IFA and shown to be in close proximity to the nucleus as dots.
