Upper: Immunofluorescence assay using the Pfμ1-specific antibody. P. falciparum 3D7 schizont/merozoite stage parasites were immunostained with antibodies specific to the Pfμ1 and rhoptry-localized RAP1 protein. The parasite nuclei were stained with DAPI and the slides were visualized by confocal laser scanning microscopy. Scale bars denote 5 μm. Lower: Immunofluorescence assay to study relative localization of Pfμ1 and RAP1 in trophozoite and schizont stages.
Lower: Transgenic parasites expressing Pfμ1–GFP were immunostained with antibodies specific to RAP1. Pfμ1 and RAP1 showed colocalization in late trophozoite stages and early schizont stages indicating packaging of RAP1 in the AP-1 containing vesicles in earlier stages and trafficking to rhoptries in later stages of parasite development. LT, Late Trophozoite; ES, Early Schizont; LS, Late Schizont. Scale bars denote 5 μm.
