Subcellular localization of native Plasmodium falciparum surface-related antigen (PfSRA) in asexual stages. Methanol-fixed 3D7 ring, trophozoite, schizont, and released merozoites were stained with α-PfSRA antibodies. A, Anti-PfSRA P1 antibody (green; 1:100). B, Anti-PfSRA P2 antibody (green; 1:100). C, Anti-PfSRA P3 antibody (green; 1:100). Colabeling of rabbit α-PfSRA-1 (red) with mouse α-PfAMA-1 (1:100; green) antibodies in intact schizonts show that PfSRA is localized on the merozoite surface. Secondary antibodies used are Alexa 488–conjugated goat α-mouse immunoglobulin G (IgG), Alexa 568-conjugated goat α-rabbit IgG (1:200; Life Technologies). Exposure times were identical for all images of the same channel. Nuclei were stained with 4′,6-diamidino-2-phenylindole (DAPI) (blue). Nuclei were stained with DAPI (blue).
