Membrane localization of PfPLP1 was detected using IFA of untreated and BAPTA-AM-treated schizonts. Schizonts were stained with anti-PfPLP1 (green) and membrane dye (red) and visualized under confocal microscope. Three-dimensional reconstruction from z-stacks of IFA images using Imaris software is shown. PfPLP1 is localized on the RBCM of untreated schizonts but not in BAPTA-AM (B/AM)-treated schizonts, indicating that intracellular Ca2+ chelation blocks membrane localization of PfPLP1.
