Assays were performed twice and ~50 attached merozoites were assessed for each RBC genetic background. Using confocal microscopy, we found that PfAMA1 and PfRON4 were frequently co localized at the cellular interface for merozoites attached to WT control cells (~85%). In comparison, for merozoites attached to CD55-null erythrocytes, PfAMA1 and PfRON4 were co-localized at the cellular interface in only ~40% of cases. For those merozoites in which PfAMA1 and PfRON4 were colocalized at the interface, we observed that approximately half of the WT control cells had an indentation at the point of merozoite contact, suggesting the merozoites were mid invasion, whereas this was never observed for the CD55-null cells.
