Localization of PfRAP01 and PfRAP21 in asexual blood stages of P. falciparum. Immuno-[fluorescence assays of PfRAP01 (a) and PfRAP21 (b) on ring (R), early trophozoite (ET), late trophozoite (LT), early schizont (ES) and late schizont (LS) stages. The IFAs are representative of three independent experiments. In accordance with the predominant localization of RAP proteins to the mitochondria or plastid, PfRAP01 and PfRAP21 have been predicted to localize to the parasite mitochondrion. To validate this prediction, we performed anti-HA immunofluorescence assays (IFAs) to detect the 3x-HA tagged PfRAP01 and PfRAP21 proteins. We showed that PfRAP01 and PfRAP21 both co-localized with MitoTracker, a red-fluorescent dye that stains mitochondria in live parasites.
