PF3D7_1346100 protein transport protein SEC61 subunit alpha

Intracellular location of PfSec61 analysed by immunofluorescence confocal microscopy. Synchronized parasite cultures were reacted with the mouse anti-PfSec61 (diluted 1:20) (1, 2 and 3) or rabbit anti-Endoplasmic Reticulum (4, 5 and 6), revealed by FITC-conjugated goat anti-species IgG. Nuclei were labeled with propidium iodide. PfSec61 is located in the cytoplasm of ring stages (1 and 4), Trophozoites (2 and 5) and Schizonts (3 and 6). Panel 7 and 9 correspond to triple-labeled P. falciparum and C32 melanoma cells, respectively. The anti-PfSec61 was revealed with anti-mouse FITC; the anti-ER revealed with anti-rabbit Cy5; nuclei were stained with propidium iodide (colour modified to cyan). The yellow colour is due to the images superimposition demonstrating that anti-mouse PfSec61 recognises the endoplasmic reticulum. Panel 8: 5 μm scale.
