PF3D7_1148700 Plasmodium exported protein (PHISTc), unknown function gametocyte exported protein 12

Expression analysis and co-localization of PF11_0503 with PfSBP-1 and PfEMP-1 at different stages of asexual parasite life cycle. Smears of methanol fixed Pf3D7-infected erythrocytes were stained with anti-PHIST antibodies (1:500), anti PfSBP-1 antibodies (1:1000) and anti PfEMP-1 antibodies (1:500), followed by incubation with Alexa flour-conjugated secondary antibodies (Alexa fluor 488 – green and Alexa fluor 546 – red). DIC: differential interference contrast image, DAPI: nuclear staining using DAPI (blue), PfEMP-1: mice anti-ATS antibody (red), PfSBP-1: anti-PfSBP-1 antibody (red), PF11_0503 : rabbit anti-PF11_0503 antibodies (green), Merge: overlay of PHIST proteins with PfEMP-1 or PfSBP-1. PF11_0503 showed a similar expression pattern as that of PfSBP-1 at the ring stage of parasite development (C). At mature trophozoite and early schizont stages, PF11_0503 showed limited overlap with PfSBP-1 as punctuate foci, while a more homogenous distribution around the periphery of iRBCs surrounding PfSBP-1 fluorescence could be seen at mature schizont stage. This PHIST protein had a very confined and remarkable overlapping with PfEMP-1 in mature schizonts, suggesting iRBC membrane to be their final destination.
