Orc2 occupies perinuclear sites. (A) Co-IFA of Orc2 and Actin I shows a significant perinuclear colocalization of both the proteins. Actin I has been reported to be present at perinuclear sites. Ring stage parasites (18±4 hpi) were taken for this IFA (n>4). (B) Effect of cytochalasin D (10μM) on the polymerisation of Actin I and subsequent effect on Orc2 localization. PfOrc2C to co-localize with Actin I (A). Actin I has been reported to be present at the nuclear periphery in early ring stage parasites. Upon CD treatment (10μM) in ring stage parasites (18±4hpi), we found a diffused pattern of Actin I due to the loss of polymerization (B). Interestingly, we also found a considerable amount of diffusion for PfOrc2 signal.
