We observed Pfs16 signal in some parasites without detectable pigment (sexual ring), consistent with the observation that expression of this marker starts before the previously reported 30–40 h.p.i. These results suggest that single-nucleated, hemozoin-containing parasites that are positive for PfAP2-G but negative for Pfs16 are sexually committed trophozoites rather than very early stage I gametocytes that do not express Pfs16 at detectable levels yet.
