Conditional deletion of PfPATPL1-3HA exon 3 and effect on blood-stage growth. Detection of PfPATPL1-3HA in Rap+ and Rap− schizonts by IFA. Rap+ and Rap− schizonts were probed with anti-HA rat mAbs to detect PfPATPL1-3HA by IFA. PfPATPL1-3HA is completely depleted in Rap+ parasites. Nuclear DNA was stained with DNA-binding fluorescent dye H33342. IFA using anti-HA mAbs also demonstrates depletion of PfPATPL1-3HA in schizonts from rapamycin-treated cultures.
