PfATG18 participates in autophagy-like pathway in P. falciparum. Colocalisation of PfATG18 with C. PfATG8 and E. PfATG5 upon starvation for indicated time period. Immunofluorescence staining of parasites using anti-PfATG18 (1:400), anti-PfATG8 (1:600) or anti-PfATG5 (1:400) antibodies labelled using Zenon antibody labelling system. The Pearson’s coefficient (R) represents degree of colocalization between PfATG18 and PfATG8 evaluated from the signal within the square region shown in the shown in the enlarged panel, N=10 cells, n=3 experiments, Scale bar=5 μm. The number of PfATG18 puncta showing colocalisation with PfATG8 (C) and PfATG5 (E) was found to increase up to ~2-folds upon starvation. Taken together, the increase in PfATG18 expression at the mRNA as well as at the protein level and its colocalisation with PfATG8 and PfATG5 vesicles under starvation suggests that PfATG18 has a role in the autophagy-like pathway in this parasite.
