Anti-PfHU antibodies and Mitotracker Red were used as markers for apicoplast and mitochondria, respectively. DAPI (blue) in the nuclear DNA stain. Clear overlap of the apicoplast marker PfHU and PfYihA1 signal was observed (D) with no overlap with the mitochondrial dye indicating that PfYihA1 localizes exclusively in the P. falciparum apicoplast. On the other hand, PfYihA2 localized to the mitochondrion as evident from overlapping PfYihA2 signal with Mitotracker Red (E). PfYihA3 signal was distributed throughout the cytosol and also overlapped with the apicoplast marker PfHU (F). No overlap was observed with Mitotracker Red. PfYihA3 thus localizes in both the cytosol and the apicoplast.
